Ooooo I think I found it. I think they are chartering from Sun Country. They got a 737 coming in to LFT in the next couple hours and it doesn't have a flight plan out of LFT yet.

You nailed it. Your flight is scheduled for DSM. Arrives at 4:10 PM. They leave LFT at 2:30 PM.

Ames airport?
The Ames airport runway is too short for a plane large enough to bring a football team. City of Ames Airport Manager quote from 2019:

“Well, maybe one day,” Pregitzer said. “We obviously can capture any sport, basically, except for football. Football has passenger sizes where we would have to get into the commercial airport requirements. Maybe with the extended runway, we could start to get those larger charters that you see in football.”

Extending the runway is one part of several proposals presented in 2019. One of them would extend the runway from the current 5,500 feet to 8,000 feet. I do not know if any more progress has been made towards accepting/implementing a proposal -- other than I'm fairly certain no runway extension has been started. I suppose a school could charter 2 separate smaller planes but . . .

What has to happen exactly to cause a last-minute cancellation? I don't think I've seen that anywhere.
Can't field 53 players OR can't field any single position group.
I haven't seen what the position groups are, or what the minimum number is for each group, or if an athlete could count towards multiple position groups (like McDonald counting as either LB or DE).
I don't know if the 53 players/position group number must be COVID related or if it can be for any reason (grades, injuries).
I don't know if that is all of it.
I wasn't able to quickly find the definitive Big 12 protocols.
