Even for those who have read the comics, they have really veered off course in the TV show. That's part of the fun. It's the same general story line, but a lot of the details - some big details, even - are different.
The story they are on now is a LOT different. I can only make one educated guess based on the comics, so it's not really even a spoiler:

They are different but still the same. If that makes sense. Same story line arcs and mostly same people. Only difference being that events happen at different times or people.
There has already been someone cast for the Tyrese role. I think he's supposed to show up at some point this season.
I seriously doubt that. If that were the case, he would have a awfully short lifespan. If he make's an appearance, then he will have a completely different story line. Tyrese was picked up on the road with his daughter long before the prison and didn't make it out of the prison arc.
Like I said, they will probably just make up a new role for him otherwise he will just get killed off right away by the goveners men.
Likely be a lot of things different i would assume. Comic is very different from the show.
That is kinda how I see it going down. I think Merle is going to try and get Daryl on his side, but in the process, threaten the people he's formed a bond with (Rick, Carrol, etc) and he will have to be the one that puts him down. That or the Govenor does it because it's obvious he really doesn't care about anyone but himself.
